Can MIDI Designer send note-on note-off combinations to make a keyboard or a drumpad?

Yes! You can use message type "Note On/Note Off" or message type "Note On/Note On" (the latter sends a zero velocity note on for the touch off).

As of 1.4.2, position-sensitive drumpads can be created. See this post for the details.
